Evaluación de esferas de alginato conteniendo al bacteriófago S1 como alternativa para el tratamiento de salmonelosis en pollos (gallus gallus domesticus)

Las Enfermedades Transmitidas por Alimentos (ETA) son una de principales causas de morbi-mortalidad a nivel mundial, los serotipos no tíficos de Salmonella enterica son los principales agentes causales. Uno de los serotipos asociados mayormente a ETA es el serotipo Salmonella enteritidis presente en productos de origen avícola y al cual se le atribuye el 24.1% de los brotes de salmonelosis en humanos. En el año 2017, la Organización Mundial de la Salud (OMS) publicó un listado en el que clasifica la urgencia de investigar y desarrollar nuevos antimicrobianos para el tratamiento de microorganismos patógenos, catalogándolos en tres niveles de prioridad tomando en cuenta las infecciones provocadas y la resistencia a antibióticos reportados. Cabe destacar, que en dicho listado Salmonella spp se encuentra clasificada como prioridad elevada. La Fagoterapia ha demostrado ser una opción efectiva y costeable como tratamiento y control de afecciones producidas por bacterias patógenas. Actualmente, se investigan y desarrollan formas farmacéuticas sólidas para la administración de Bacteriófagos por vía oral, a partir de sistemas de liberación con biopolímeros como el alginato mejorando así la estabilidad a condiciones gástricas. El presente trabajo se enfocó en obtener un sistema de entrega vía oral capaz de brindar protección al Bacteriófago S1 tras su paso por el tracto gastrointestinal de pollos (Gallus gallus domesticus) de un mes de edad. Por ello, el Bacteriófago S1 se encapsuló con dos alginatos comerciales a una concentración del 2% p/v utilizando solución de CaCl2 al 2% p/v como agente gelificante, para cada uno de los alginatos se determinó la eficacia de encapsulación y el rendimiento fágico, posteriormente, se seleccionó el alginato 1 por mostrar mayor eficacia de encapsulación. Se procedió a realizar los ensayo in vitro correspondientes a la liberación del Bacteriófago S1 a pH intestinal (6.9) por la técnica de agitación continua, se determinó que la esfera de alginato permite la liberación del Bacteriófago S1 a partir de las tres horas de agitación, por otro lado, en los ensayos in vivo se corroboró a partir de la presencia de zonas líticas que las esferas de alginato permiten la entrega del Bacteriófago S1 en duodeno y ciegos (compartimentos que coloniza Salmonella. enteritidis), además la infección por Salmonella enteritidis se vio reducida a partir de las 48 h de administrado el tratamiento.

Currently, solid pharmaceutical forms for the administration of Bacteriophages by the oral route are investigated and developed, using release systems with biopolymers such as alginate thus improving stability to gastric conditions. The aim of this study was to obtain an oral delivery system efficient of providing protection to Bacteriophage S1 after passing through the gastrointestinal tract of one-month-old chickens (Gallus gallus domesticus). The Bacteriophage S1 was encapsulated with two commercial alginates at a concentration of 2% w / v using 2% CaCl 2 solution w / v as a gelling agent, for each of the alginates the encapsulation efficiency and the phage yield were determined, subsequently, alginate 1 was selected for showing greater encapsulation efficiency. The in vitro tests corresponding to the release of Bacteriophage S1 at intestinal pH (6.9) were carried out by the continuous agitation technique, it was determined that the alginate sphere allows the release of Bacteriophage S1 after three hours of agitation. The in vivo tests, it was corroborated from the presence of lytic zones that the alginate spheres allow the delivery of Bacteriophage S1 in the duodenum and caecum (compartments colonized by Salmonella enteritidis), besides the infection by Salmonella enteritidis It was reduced as of 48 h after the treatment was administered.
