Caracterización de aislados bacterianos patógenos a Dactylopius opuntiae (hemíptera: dactylopiidae) cochinilla silvestre del nopal en el estado de Morelos

El objetivo de este estudio fue aislar y seleccionar cepas bacterianas patógenas a Dactylopius opuntiae (Cockerell) (Hemiptera: Dactylopiidae), una plaga de Opuntia ficus-indica (L.) Mill. (Cactaceae) comúnmente conocida como “cochinilla silvestre del nopal” para proponer agentes alternativos de control ecológico contra el insecto. Cuarenta y una cepas de bacterias formadoras de esporas fueron aisladas de cadáveres de adultos y ninfas de D. opuntiae colectados en cuatro localidades diferentes del estado de Morelos, México. Los bioensayos de patogenicidad y virulencia se realizaron mediante la aplicación directa del complejo espora-cristal formados en la fase estacionaria alcanzada a las 72 horas en el medio de cultivo HCT. En los bioensayos de patogenicidad, la cepa HH41 causó 30.78% de mortalidad en ninfas del segundo estadio a las 96 horas a una concentración de 100 μg/mL de proteína total, mientras que la cepa HH10 mostró una mortalidad del 62.52% a 450 μg/mL bajo las mismas condiciones. La virulencia se obtuvo solo de la cepa HH10, que muestra un valor de CL50 de 403.293 μg/mL de proteína total. Los postulados de Köch se llevaron a cabo para corroborar que las cepas bacterianas eran los agentes patógenos de D. opuntiae. Las cepas HH10 y HH41 fueron identificadas molecularmente como Bacillus thuringiensis amplificando el gen 16S rRNA y gyrB. Se determinó, mediante la prueba de reducción de la tensión superficial y prueba de hemolisis, que las cepas HH10 y HH41 producen un biosurfactante (BS) los BS son moléculas anfifílicas que han sido reportadas con actividad insecticida. Para medir la tensión superficial se realizaron bionensayos (TS) con los sobrenadantes, lo que redujo el TS del agua de 72 mD /m a 55.3 para la cepa HH10 y a 53.2 para la cepa HH41. Finalmente, se realizaron pruebas de hemólisis para ambas cepas bacterianas, que fueron positivas.

The objective of this study was to isolate and select pathogenic bacterial strains to propose an ecological alternative to control the insect Dactylopius opuntiae (Cockerell) (Hemiptera: Dactylopiidae), a pest of Opuntia ficus-indica (L.) Mill. (Cactaceae) commonly known as “wild cochineal of the nopal”. Forty-one spore- forming bacterial strains were isolated from corpses of adults and nymphs of D. opuntiae collected in four different locations in the state of Morelos, Mexico. The bioassays were performed by direct application of the spore-crystal complex formed in the stationary phase reached after 72 hours, the strains were cultivated in HCT medium. In the pathogenicity bioassays, the HH41 strain showed a mortality of 30.78 % for seconds instar nymphs after 96 hours at 100 μg / mL of total protein, while the HH10 strain showed a mortality of 62.52% at 450 μg / mL under the same conditions. Virulence was obtained only the HH10 strain, which showed an LC50 value of 403.293 μg / mL of total protein. Köch's postulates were carried out to corroborate that the bacterial strains applied were pathogenic to D. opuntiae. Strains HH10 and HH41 were molecularly identified to species level by amplification of the by 16S rRNA and gyrB genes as Bacillus thuringiensis. The production of a surfactant by the HH10 and HH41 strains was determined by a surface tension reduction test and hemolysis test, the BS are amphiphilic molecules that have been reported with insecticidal activity. Bioassays using the cell free, wasted culture media (CFM) were carried out in order to measure the effect on water surface tension (TS). HH10 and HH41 CFMS reduced the water from 72 to 55.3 mD/m and 72 to 53.2 mD/m respectively. The two strains showed positive hemolitical activity in tests performed using erythrocites.
