Herramientas moleculares para el estudio de las hidrofobinas SIH1, SIH2 y SIH4 del hongo halófilo Aspergillus sydowii

Resumen Los halófilos son aquellos organismos que crecen de manera óptima en presencia de NaCl (≥200mM), mientras que los halotolerantes son aquellos que pueden crecer en condiciones de alta salinidad, pero su crecimiento óptimo es en su ausencia. Ambos grupos de organismos son relevantes porque pueden servir como fuente de biomoléculas tales como enzimas estables en alta fuerza iónica, biopolímeros y solutos compatibles, además de ser útiles para procesos de biofermentación, y algunos son candidatos potenciales para la biorremediación. La mayor parte de los organismos halófilos caracterizados hasta la fecha son procariontes, aunque se han descrito algunas algas y hongos con la potencialidad de crecer en condiciones de altas concentraciones de NaCl (>1.0M). Dentro de los mecanismos de halofilia observados en hongos están: el aumento de transportadores de membrana, la acumulación de solutos compatibles y el engrosamiento de la pared celular. Un nuevo mecanismo propuesto es la expresión diferencial de hidrofobinas respecto a la salinidad del medio, lo que se ha visto en el halófilo extremo Wallemia ichthyophaga (Zajc et al., 2013). En el análisis transcriptómico del hongo Aspergillus sydowii BHH004, realizado en nuestro laboratorio, se observó la expresión diferencial de tres hidrofobinas, SIH1 y SIH2 en presencia de 2 M de NaCl y SIH4 en la condición sin sal (Pérez-Llano et al., 2020). Dado que existen pocos estudios que aborden el papel de las hidrofobinas en la halofilia, resulta relevante el análisis de las hidrofobinas en varios niveles: in silico, in vitro e in vivo. Por tanto, el objetivo del presente trabajo fue generar herramientas para el estudio de las hidrofobinas SIH1, SIH2 y SIH4 del hongo halófilo Aspergillus sydowii. Los resultados in silico mediante la generación de modelos tridimensionales nos indican que estas proteínas tienen una carga electrostática negativa, que se encuentra localizada en una de sus caras, aunque SIH4 tiene mayor homogeneidad de cargas negativas que las otras dos. Para estudiar las hidrofobinas in vitro, hemos generado las construcciones pPICZ::SHI1, pPICZ::SIH2 y pPICZ::SIH4 para la posterior expresión en el sistema heterólogo Pichia pastoris y purificación; para hacer estudios biofísicos de su comportamiento a distintas concentraciones de sal. Para los estudios in vivo se realizaron las construcciones para el uso del sistema CRISP/Cas, sin embargo, los RNA guías no fueron introducidos de manera eficiente en el vector, por lo que generamos nuevos oligonucleótidos para hacer las construcciones de manera tradicional por recombinación homóloga. A manera de conclusión, los análisis in silico mostraron que las hidrofobinas SIH1, SIH2 y SIH4 de manera general son proteínas ácidas. Se generaron las construcciones para la expresión heteróloga de las hidrofobinas en Pichia pastoris. No se obtuvieron las construcciones adecuadas para generar a las cepas mutantes de estas proteínas. En estudios a mediano y largo plazo se generarán las cepas mutantes a través del sistema de recombinación homóloga, y se expresarán y purificarán las hidrofobinas para su posterior análisis biofísico.

Abstract Halophiles are organisms that grow optimally in the presence of NaCl (≥200mM), while halotolerants can grow in hypersaline conditions, but their optimal growth is in the absence of salt. Both groups are relevant because they can serve as a source of biomolecules such as stable enzymes in high ion strength, biopolymers, and compatible solutes, as well as being useful for biofermentation processes, and some are potential candidates for bioremediation. Most of the halophilic organisms characterized to date are prokaryotes, although some algae and fungi have been described with the potential to grow in hypersaline conditions (NaCl >1.0M). Among the halophile mechanisms observed in fungi we can find increased number of membrane transporters, accumulation of compatible solutes, and thickening of the cell wall. A new mechanism proposed is the differential expression of hydrophobins regarding the salinity of the medium, which has been seen in the extreme halophile Wallemia ichthyophaga (Zajc et al., 2013). In the transcriptomic analysis of the fungus Aspergillus sydowii BHH004, carried out in our laboratory, the differential expression of three hydrophobins, SIH1 and SIH2 were observed in the presence of 2M of NaCl and SIH4 in the condition without salt (Pérez-Llano et al., 2020). Since there are few studies addressing the role of hydrophobins in halophilia, the analysis of hydrophobins at various levels is relevant: in silico, in vitro, and in vivo. Therefore, the objective of this work was to generate tools for the study of SIH1, SIH2, and SIH4 hydrophobins of the halophilic fungus Aspergillus sydowii. The results at the in-silico level by generating three-dimensional models indicate that these proteins have a negative electrostatic charge, that is located on one of their faces, although SIH4 has greater homogeneity of negative charges than the other two. To study hydrophobins in vitro, we have generated the constructions pPICZ::SHI1, pPICZ::SIH2 y pPICZ::SIH4 for further expression in the Pichia pastoris heterologous system and purification, to do biophysical studies of their behavior at different salt concentrations. For in vivo studies, constructions were carried out for the use of the system CRISP/Cas, however, the guide RNAs were not efficiently introduced into the vector, so we generated new oligonucleotides to make the constructions in a traditional way by homologous recombination. To summarize, in silico analyses showed that SIH1, SIH2, and SIH4 hydrophobins are generally acidic proteins. Constructions were generated for the heterologous expression of hydrophobins in Pichia pastoris. The right constructions were not obtained to generate the mutant strains of these proteins. In medium and long-term studies, the mutant strains will be generated through the homologous recombination system, and the hydrophobins will be expressed and purified for their subsequent biophysical analysis.
