El cáncer de pulmón es la primera causa de muerte, por cáncer, en el mundo. El diagnóstico
y tratamiento de la enfermedad es tardío y el nivel de respuesta al tratamiento es pobre. El
cáncer se origina a partir del daño al ADN, esto provoca una pérdida en la regulación del
ciclo celular, inhibición de la apoptosis y proliferación incontrolada. Estudios previos han
demostrado que la anfotericina B (AmB) tiene efectos antineoplásicos y potencia el efecto
de otros antineoplásicos. L-Histidine Amida Derivada de AmB (A21) es un derivado de AmB
desarrollado por académicos de la UAEM, la UNAM y el CINVESTAV. Estudios preclínicos
previos han demostrado que el A21 tiene la misma eficacia antimicótica y es menos tóxico
que la AmB. Por lo cual, es importante saber si el derivado produce los mismos efectos
antineoplásicos que su precursor. Se usaron células de carcinoma de pulmón de células no
pequeñas (A549). Las células Se trataron con: cisplatino, carboplatino, AmB, A21,
cisplatino+AmB, cisplatino+A21, carboplatino+AmB, carboplatino+AmB. Se evaluó la
citotoxicidad y proliferación celular mediante la técnica MTT. Se identificó el tipo de muerte
celular mediante la tinción con bromuro de etidio y naranja de acridina. Se evaluó el daño
al ADN (HSC DNA Damage kit) y el efecto sobre la capacidad invasiva celular. De cada
fármaco individual se obtuvieron concentraciones inhibitorias 25 (IC25). Las IC25
encontrados fueron: cisplatino 10, carboplatino 515, AmB 29 y A21 67 μg/ml, estas
concentraciones se usaron para hacer las combinaciones. En la evaluación de la
citotoxicidad y proliferación se observó que la combinación cisplatino+A21 redujo un 40 y
70% aproximadamente la viabilidad y la tasa de proliferación respectivamente, mientras que
el tratamiento carboplatino+A21 redujo un 50 y 95% aproximadamente, por otro lado, el
tratamiento A21 redujo un 40% la tasa de proliferación comparado con el control. Se
identifico que los tratamientos A21, cisplatino+A21 y Carboplatino+A21 indujeron muerte
por apoptosis aproximadamente en un 45, 75 y 80% de células, respectivamente. En el
ensayo de invasión se obtuvo que los tratamientos A21, cisplatino+A21 y Carboplatino+A21
redujeron un 40, 40 y 70% la invasión de células, en comparación al control positivo. El
derivado A21 fue más e igual de efectivo que la AmB para inhibir invasión celular e inducir
apoptosis, respectivamente. El tratamiento cisplatino+A21 produjo mayor efecto citotóxico,
anti-proliferativo, genotóxico e indujo mayor apoptosis que el derivado solo, pero menos
efecto citotóxico que cisplatino+AmB. El tratamiento carboplatino+A21 fue más efectivo que
la combinación de carboplatino+AmB, y que el A21 solo y combinado con cisplatino. Con
base a los resultados obtenidos se puede concluir que el derivado A21 tiene efectos
antitumorales y produce un efecto sinergista con cisplatino y carboplatino en células A549. | es_MX |

Lung cancer is the leading cause of death, due to cancer, in the world. The diagnosis and
treatment of the disease is late and the level of response to treatment is poor. Cancer
originates from DNA damage, this causes a loss in cell cycle regulation, inhibition of
apoptosis and uncontrolled proliferation. Previous studies have shown that amphotericin B
(AmB) has antineoplastic effects and potentiates the effect of other antineoplastic drugs. LHistidine
Amide Derivative from AmB (A21) is a derivative of AmB developed by academics
of the UAEM, UNAM and CINVESTAV. Previous preclinical studies have shown that A21
has the same antifungal efficacy and is less toxic than AmB. Therefore, it is important to
know if the derivative produces the same antineoplastic effects as its precursor. Non-small
cell lung carcinoma cells (A549) were used. Cells were treated with: cisplatin, carboplatin,
AmB, A21, cisplatin + AmB, cisplatin + A21, carboplatin + AmB, carboplatin + AmB.
Cytotoxicity and cell proliferation were evaluated using the MTT technique. The type of cell
death was identified by staining with ethidium bromide and acridine orange. DNA damage
(HSC DNA Damage kit) and the effect on cellular invasive capacity were evaluated. In each
individual drug, 25 inhibitory concentrations (IC25) were obtained. The IC25 found were:
cisplatin 10, carboplatin 515, AmB 29 and A21 67 μg / ml, these concentrations were used
to make the combinations. In the evaluation of cytotoxicity and proliferation it was observed
that the cisplatin+A21 combination reduced approximately 40 and 70% approximately
viability and proliferation rate respectively, while carboplatin+A21 treatment reduced
approximately 50 and 95%, on the other hand, the A21 treatment reduced the proliferation
rate by 40% compared to the control. It was identified that treatments with A21,
cisplatin+A21 and Carboplatin+A21 induced apoptotic death in approximately 45, 75 and
80% of cells, respectively. In the invasion test it was obtained that treatments A21,
cisplatin+A21 and Carboplatin+A21 reduced cell invasion by 40, 40 and 70%, compared to
the positive control. The A21 derivative was more and equally effective than the AmB to
inhibit cell invasion and induce apoptosis, respectively. The cisplatin+A21 treatment
produced a greater cytotoxic, anti-proliferative, genotoxic effect and induced greater
apoptosis than the derivative alone, but less cytotoxic effect than cisplatin+AmB. The
carboplatin+A21 treatment was more effective than the combination of carboplatin+AmB,
and that the A21 alone and combined with cisplatin. Based on the results obtained, it can
be concluded that derivative A21 has antitumor effects and produces a synergistic effect
with cisplatin and carboplatin in A549 cells. | es_MX |
