Cernimiento biológico de algunas especies vegetales del género ageratina

RESUMEN Los conocimientos etnomédicos en la medicina tradicional mexicana, han permitido identificar plantas con propiedades antimicrobianas, tal es el caso de las plantas conocidas comúnmente como “axihuitl” (Monroy y astillo 2010) pertenecientes al género Ageratina antes Eupatorium (familia Asterácea). Las especie vegetales pertenecientes al género Ageratina, generalmente se emplean en la medicina tradicional para el tratamiento de infecciones en la piel e infecciones gastrointestinales, así como en lavado de heridas (Cragg y Newman, 2013; Henrich et al., 2013; López y Gutiérrez, 2009). Por ello, el objetivo de este trabajo fue determinar el potencial farmacológico (antimicrobiano, antiinflamatorio y antioxidante) de tres especies del género Ageratina conocidas en la medicina tradicional de manera indistinta como “axihuitl”. Las especies del género Ageratina recolectadas fueron: A. aschemborniana (30224) del municipio de Tepoztlán; A. glabrata (30225) del municipio de Tlalnepantla y A. pazcuarensis (30226) del municipio de Huitzilac. Las tres diferentes especies de Ageratina, se sometieron a un proceso de maceración (partes aéreas) utilizando diferentes disolventes de polaridad creciente (hexano, acetona y metanol). Posteriormente, mediante presión reducida se obtuvieron tres extractos orgánicos para cada especie, a los cuales se les realizó un cernimiento de tres actividades biológicas: antimicrobiana (método de microdilución en placa), antiinflamatoria (modelo murino de edema auricular inducido con TPA) y capacidad antioxidante (ABTS, DPPH y CARF). El extracto hexánico de A. glabrata presentó el mayor potencial antimicrobiano sobre los microorganismos probados que fueron: P. aeruginosa ( M 250 μg/mL), K. pneumoniae ( M 250 μg/mL), P. mirabilis ( M 250 μg/mL), E. coli (CMI 250 μg/mL), S. aureus ( M 125 μg/mL), C. albicans ( M 32 μg/mL) y C. glabrata (CMI >1000 μg/mL), además demostró tener un amplio espectro contra bacterias Gram positivas y Gram negativa. Sin embargo los extractos de A. aschemborniana fueron más especifico contra levaduras (CMI de 4 μg/mL contra C. albicans y 32 XIV μg/mL contra C. glabrata). Por otro lado, las tres especies de Ageratina presentaron actividad antiinflamatoria; extracto metanol de A. aschemborniana con 80.36%, extracto acetónico de A. glabrata con 87.24% y extracto acetónico A. pazcuarensis con 89. 63 % de inhibición inflamatoria, teniendo como control indometacina con un 91.80%, todas las muestras fueron probadas a una concentración de 0.1 mg/oreja. En el caso de la actividad antioxidante, se observó que el extracto acetónico de A. aschemborniana fue el más activo (CI50 = 6.51μg/mL) por el método A ; sin em argo comparado contra el control positivo (extracto estandarizado de C. sinensis) la actividad fue de 50 %. Los resultados del presente trabajo contribuyen a la generación de evidencia cientifica que permite validar el uso tradicional de las especies del género Ageratina estudiadas, asi como una posible fuente de obteción de principios bioactivos, para contribuir al cuidado de la salud.

ABSTRACT The ethnomedical knowledge in Mexican traditional medicine allowed us to find plants with antimicrobial properties, such as the plants commonly known as "axihuitl" (Monroy and Castillo, 2010), belonging to the genus Ageratina before Eupatorium (Asteraceae family). Plant species belonging to the genus Ageratina are generally used in traditional medicine for the treatment of skin and gastrointestinal infections as well as wound washing. (Cragg and Newman, 2013, Henrich et al., 2013, López and Gutiérrez, 2009). Therefore, the objective of this work was to determine the pharmacological potential (antimicrobial, anti-inflammatory and antioxidant) of three species of the genus Ageratina known in traditional medicine indistinctly as "axihuitl". Ageratina aschemborniana (30224) was colleted in the town of Tepoztlán, A. glabrata (30225) was found in Tlalnepantla and A. pazcuarensis (30226) was obtained in Huitzilac. These three species of Ageratina were subjected to a maceration (aerial parts) process using different solvents of increasing polarity (hexane, acetone and methanol). Subsequently, three crude organic extracts were obtained for each species, which were monitored for three biological activities: antimicrobial (plate microdilution method), antiinflammatory (murine model of atrial edema induced with TPA) and antioxidant capacity (ABTS, DPPH and CARF). The hexane extract of A. glabrata showed the highest antimicrobial potential on the microorganisms tested: P. aeruginosa (M 250 μg/mL) K. pneumoniae (M 250 μg/mL) P. mirabilis (M 250 μg/mL). ) E. coli (M 250 μg/mL) S. aureus (M 125 μg/mL) C. albicans (M 32 μg/mL) and C. glabrata (MIC> 1000 μg/mL) also proved to have a broad spectrum against Gram positive and Gram negative bacteria. However, the extracts of A. aschemborniana were more specific against yeasts (M of 4 μg/mL against C. albicans and 32 μg/mL against C. glabrata). On the other hand, the three species of Ageratina presented anti-inflammatory activity; methanol extract of A. aschemborniana with 80.36%, acetonic extract of A. glabrata with 87.24% and acetonic extract A. pazcuarensis with 89. 63% of XVI inflammatory inhibition, taking control of indomethacin with 91.80%, all the samples were tested at a concentration of 0.1 mg / ear. In the case of antioxidant activity, it was observed that the acetonic extract of A. aschemborniana was the most active ( 50 = 6.51μg / mL) y the A TS method; however, compared to the positive control (standardized extract of C. sinensis) the activity was 50%. The results of this work contribute to the generation of scientific evidence that allows to validate the traditional use of the Ageratina species studied, as well as a possible source of bioactive principles, to contribute to health care.
