Producción y formulación de trichoderma asperellum para el manejo de patógenos de la raíz de caña de azúcar

La caña de azúcar es uno de los cultivos más representativos del estado de Morelos abarcando una superficie de 22 128 has con un rendimiento promedio de 111 ton/ha. La producción es afectada por varios factores, uno de mayor importancia son los hongos del genero Fusarium spp. que infectan las raíces lo cual ocasiona la marchitez de las plantas. Habitualmente para el control de esta enfermedad se recomienda la aplicación de diferentes fungicidas cuya efectividad es limitada y ocasionan daños al ambiente, por lo que es necesario buscar alternativas viables para el manejo de esta enfermedad. Como resultado de investigaciones anteriores en el Centro de Investigaciones Biológicas de la UAEM se ha integrado un cepario que cuenta con aislamientos de Trichoderma spp., obtenidos de campos de cultivo de caña de azúcar de varios municipios del estado de Morelos. Mediante pruebas in vitro se ha demostrado que la cepa de T. asperellum es altamente antagonista a Fusarium spp. Este trabajo tuvo como objetivo general evaluar la producción de conidios de T. asperellum en dos sustratos naturales y proponer una formulación para su aplicación en campo. Se evaluarón los sustratos de granos de arroz y sorgo con diferente contenido de humedad con tiempos variados de esterilización, bajo un diseño completamente al azar con 16 tratamientos, un testigo y cuatro repeticiones para cada sustrato. Las variables respuesta fueron: la producción de conidios y la cantidad de biomasa (mezcla de residuos del sustrato y conidios). Asimismo, se determinó la cantidad óptima de inoculo de T. asperellum para la producción masiva de conidios en sustrato de arroz. La mayor producción de conidios (9.34 X 108 conidios/g) se presentó en el sustrato de arroz con un contenido de humedad de 28.58% esterilizado durante 15 minutos y la mayor cantidad de biomasa se obtuvo con el sustrato con un contenido de humedad del 29.44% y fue esterilizado durante 15 minutos (P≤0.05, Tukey). En el sustrato de sorgo se registró la mayor producción de conidios/g (1.49 X 108) cuando presentó una humedad de 20.87% y fue esterilizado por 15 minutos (P<0.05); sin embargo, el análisis de varianza para la cantidad de biomasa no mostró diferencias significativas entre los tratamientos, presentando menos de 0.56 g. Para la producción masiva de conidios de T. asperellum sobre sustrato de arroz la cantidad óptima 2 de inóculo fue de 3 mL. Se evaluaron tres formulaciones compuestas con talco, yeso y biomasa de T. asperellum, bajo un diseño completamente al azar con cuatro tratamientos incluido un testigo y cuatro repeticiones. La viabilidad de las formulaciones se determinó mediante la evaluación de la germinación de los conidios en cada una de las formulaciones y en el testigo, al establecer las formulaciones y a intervalos de 30 días por 3 meses almacenados en oscuridad y a temperatura ambiente; también, se determinó la cantidad de UFC/g de cada formulado. En las formulaciones no se observó una disminución progresiva de la viabilidad de los conidios y la cantidad de UFC en los tres meses de almacén. En la evaluación inicial de las formulaciones 1, 2, 3 y el testigo presentarón 92.83, 90.57, 89.20 y 88.94% de viabilidad, respectivamente. Esta viabilidad se redujo gradualmente después de 3 meses de almacén a 92.31, 88.22, 89.09 y 87.50% respectivamente. La cantidad de UFC/g de los formulados y el testigo se redujo en un 0.53, 2.63, 0.13 y 1.6% durante los tres meses de almacén.

Sugarcane is one of the most representative crops in the state of Morelos covering an area of 22 128 ha with an average yield of 111 ton/ha. The production is affected by several factors, one of major importance are the fungi of the genus Fusarium spp. that infect the roots causing a wilting of the plants. Usually for the control of this disease is recommended the application of different fungicides whose effectiveness is limited and cause damage to the environment, so it is necessary to look for viable alternatives for the management of this disease. As a result of previous investigations in the Center of Biological Research of the UAEM has been integrated a strains collection that counts with isolates of Trichoderma spp., obtained from fields of sugarcane of several municipalities of the state Morelos. In vitro tests have shown that the T. asperellum strain is highly antagonistic to Fusarium spp. The general objetive of this work was evaluate the production of conidia of T. asperellum in two natural substrates and to propose a formulation for its application in field. Substrates of grains of rice and sorghum with different moisture content and with varying sterilization times were evaluated, under a completely random design with 16 treatments, one control and four repetitions for each substrate. The response variables were: the production of conidia and the amount of biomass (mixture of substrate residues and conidia). The optimal amount of T. asperellum inoculum was also determined for the massive production of conidia in rice substrate. The highest production of conidia (9.34 X 108 conidia/g) was present in rice substrate with a moisture content of 28.58% sterilized for 15 minutes and the highest amount of biomass was obtained with the substrate with a moisture content of 29.44% and sterilized for 15 minutes (P≤0.05, Tukey). In the sorghum substrate the highest conidia production (1.49 X 108 conidia/g) was registered when it present a humidity of 20.87% and was sterilized during 15 minutes (P<0.05); however, the analisis of variance for the amount of biomass showed no significant difference between treatments, presenting less than 0.56 g. For the massive production of T. asperellum conidia on rice substrate the optimal amount of inoculum was 3 mL. Three formulations composed of talc, gypsum and T. asperellum biomass were evaluated under a completely randomized desing with four treatments including a control and four repetitions. 4 The viability of the formulations was determined by evaluating the germination of the conidia in each of the formulations and in the control, when establishing the formulatins and at intervals of 30 days for 3 months stored in the dark and at room temperatura; the amount of CFU/g of formulated was also determined. In the formulations there was no a progressive decrease in the viability of the conidia and the amount of CFU in the three months of storage. In the initial evaluation of formulations 1, 2, 3 and the control presented 92.83, 90.57, 89.20 and 88.94% viability, respectively. This viability was gradually reduced after 3 months of storage at 92.31, 88.22, 89.09 and 87.50% respectively. The amount of CFU/g of the formulated and the control was reduced by a 0.53, 2.63, 0.13 and 1.6% during the three months of storage.
