dc.descriptionRESUMEN En condiciones de bajo potencial de agua el crecimiento de la raíz de plántulas de Arabidopsis thaliana de tipo silvestre (WT) se abate. Sin embargo, el crecimiento de la raíz de la mutante altered hydrotropic response1 (ahr1) de A. thaliana, en estas mismas condiciones, es similar a lo observado en plántulas crecidas en condiciones control (sin disminución del potencial de agua). En este trabajo, con la finalidad de determinar la relación de estos parámetros con el hidrotropismo, se utilizaron diferentes métodos para determinar los cambios en los procesos celulares en la raíz, así como de los componentes responsables de mantener la elongación radicular en condiciones de bajo potencial de agua. Los análisis celulares del meristemo apical de la raíz (RAM, del inglés “root apical meristem”) y de la zona de elongación (ZE) de la raíz de la mutante ahr1, crecida en el sistema WPGS (del inglés, “water potential growth system”), desarrollado en el presente trabajo, mostraron un aumento de las células tanto del meristemo, como de la zona de elongación, células de la corteza completamente elongadas y una duración del ciclo celular, acordes al crecimiento en condiciones normales. Es decir, bajo condiciones de déficit hídrico, el crecimiento de las raíces de la mutante ahr1 no sufre alteraciones, lo cual se explica por su capacidad de mantener tanto la producción, como la elongación celular al mismo nivel en condiciones control y de déficit hídrico. Un examen de la respuesta a auxina, mediante el sistema reportero DR5::GUS, indicó que la señalización de auxinas participa en la disminución del crecimiento de las raíces de WT en condiciones de bajo potencial de agua en tanto que en las raíces de la mutante ahr1 presentan una baja respuesta a este regulador del crecimiento vegetal. Además, dado que en algunas plantas, como A. thaliana, uno de los mecanismos para contender con el déficit hídrico es la acumulación de osmolitos,, como la prolina tanto en hojas como en raíces, se comparó la acumulación de la prolina en raíces de plantas de tipo silvestre y de la mutante ahr1. Las raíces de las plantas WT en condiciones de bajo potencial de agua mostraron una mayor cantidad de prolina, que las de la mutante ahr1. Finalmente, la expresión del gen D1-6 pirrolina-5-carboxilato sintetasa (P5CS1), implicado en la síntesis de prolina, es mucho mas alta en plántulas WT en comparación con la mutante ahr1. En conjunto, estos resultados revelaron un fenotipo de la mutante ahr1 que representa un importante recurso para tratar de entender la señalización y mecanismos de control del hidrotropismo.es_MX
dc.descriptionABSTRACT Under conditions of low water potential, root growth of wild type seedling Arabidopsis thaliana (WT) is abated. However, the growth of the root of the altered the hydrotropic response1 (ahr1) mutant of A. thaliana, in the same conditions, is similar to that observed in seedlings grown under control conditions (no decrease in water potential). In this work, in order to determine the relationship of these parameters with hydrotropism, different methods were used to determine changes in cellular processes of the root, as well as the components responsible for maintaining root elongation in conditions of low water potential. The cellular analysis of the root apical meristem (RAM) and of the elongation zone of roots of the ahr1 mutant, grown in the WPGS system (water potential growth system) developed in the present work, resulted in an increase of the cells both in meristem, as well as in the elongation zone, in the completely elongated cortex cells and in the cell cycle duration, according to the growth under normal conditions. That is, under conditions of water deficit, the growth of roots of the ahr1 mutant did not suffer alterations, which is explained by its ability to maintain both production and cell elongation at the same level under control and water deficit conditions. An examination of the response to auxin, using the reporter system DR5::GUS, indicated that auxin signaling participates in the reduction of the growth of WT roots in conditions of low water potential, while in roots of the ahr1 mutant presented a low response to this plant growth regulator. In addition, since in plants one of the mechanisms to contend with water deficit is the accumulation of osmolytes (such as proline), both in leaves and roots, the accumulation of proline in roots of WT plants was compared of the ahr1 mutant. The roots of WT plants in conditions of low water potential showed a greater amount of proline, than those of the ahr1 mutant. Finally, the expression of the D1-6 pyrroline-5-carboxylate synthetase gene (P5CS1), involved in the synthesis of proline, is considerably higher in WT seedlings than in those of the ahr1 mutant. Taken together, these results revealed that the phenotype of the ahr1 mutant represents an important resource for understanding the signaling and control mechanisms of hydrotropism.es_MX
