dc.description | RESUMEN
El poliovirus es uno de los agentes infecciosos en vías de erradicación a nivel global,
actualmente Afganistán y Pakistán son los únicos países en donde el virus se mantiene
endémico (serotipo 1); sin embargo, se han reportado rebrotes de poliomielitis ocasionados
por poliovirus derivado de vacuna circulante (cVDPV) en países donde el virus no es
considerado endémico. La Organización Mundial de la Salud recomienda mantener elevados
los niveles de cobertura de vacunación sistemática, así como buscar nuevas alternativas de
vacunación efectiva y accesible para combatir infecciones provocadas por poliovirus.
En los últimos años se han desarrollado técnicas para el aprovechamiento de cultivos
vegetales con capacidad de producir vacunas a través de proteínas recombinantes. Una de las
ventajas de los productos biofarmacéuticos a partir de plantas es que, al ser administradas de
forma oral, ayudan a la producción de una respuesta inmunitaria en la mucosa. Daucus carota
es una de las especies con gran oportunidad en este campo biotecnológico, dado que produce
tejido que es de uso comestible en todo el mundo por lo cual se considera como un recurso
de importancia para la producción de vacunas comestibles.
Rosales-Mendoza en el año 2018 realizó la transformación y obtención de células de
zanahoria que expresan la proteína VP2 que junto con VP1, VP3 y VP4 conforman la cápside
del poliovirus, de gran importancia para el desarrollo de una vacuna. En el presente trabajo
se estableció la producción de cultivos celulares de Daucus carota in vitro de callos y células
en suspensión, con la capacidad de producir proteína VP2, con relevancia en la inducción de
respuesta inmune contra poliovirus.
A partir de los subcultivos de callos de la línea transformada (C-1) y de la línea silvestre (CWT) se obtuvieron callos friables bajo tres condiciones de crecimiento (luz constante,
fotoperiodo y oscuridad) y seis tratamientos de medio de cultivo.
Los resultados obtenidos en este proyecto de investigación permitieron el establecimiento
del cultivo in vitro de Daucus carota de la línea SMC-1 lo que representa el primer reporte
de células en suspensión establecidas en matraz para la producción de una vacuna contra
poliovirus. Así mismo, se muestran los resultados obtenidos en una cinética de crecimiento
celular de la línea SMC-1, se registró un máximo de biomasa es 16.07 g L-1 PS a los 12 días
con una velocidad de crecimiento de 0.2938 d-1
y un tiempo de duplicación de 2.35 días lo

Currently, Afghanistan and Pakistan are the only countries where poliovirus remains endemic
(serotype 1); however, polio outbreaks caused by circulating vaccine-derived poliovirus
(cVDPV) have been reported in countries where the virus is not considered endemic. The
World Health Organization recommends maintaining high levels of routine vaccination
coverage, as well as seeking new effective and accessible vaccination alternatives to combat
poliovirus infections.
In recent years, techniques have been developed for the use of plant crops with the capacity
to produce vaccines through recombinant proteins. One of the advantages of plant-based
biopharmaceuticals is that, when administered orally, they help to produce an immune
response in the mucosa. Daucus carota is one of the species with great opportunity in this
biotechnological field, since it produces tissue that is of edible use all over the world or for
which it is considered as an important resource to produce edible vaccines.
In 2018, Rosales-Mendoza carried out the transformation and obtention of carrot cells that
express the protein (VP2); that together with VP1, VP3 and VP4 make up the capsid of the
poliovirus, of great importance for the development of a vaccine. In the present work, the
production of in vitro Daucus carota cell cultures of callus and suspension cells, with the
ability to produce VP2 protein, with relevance in the induction of immune response against
poliovirus, was established.
From callus subcultures of the transformed line (C-1) and the wild-type line (WT), friable
callus was obtained under three growth conditions (constant light, photoperiod and darkness)
and six culture medium treatments.
The results obtained in this research project allowed the establishment of the in vitro culture
of Daucus carota of the SMC-1 line, which represents the first report of cells in suspension
established in a flask for the production of a vaccine against poliovirus. The results obtained
in the cell growth kinetics of the SMC-1 line are shown; a maximum biomass of 16.07 g L-1
PS at 12 days a growth rate of 0.2938 d-1
and a doubling time of 2.35 days were recorded,
which allow generating an interesting cell culture for the realization of new projects with
other culture strategies that allow improving the biomass yield and the production of the VP2
