En el continente americano los vipéridos (“víboras”) son los responsables de la
mayoría de los accidentes ofídicos, principalmente por los géneros Bothrops y Crotalus 1.
Los anticuerpos utilizados para los antivenenos con fines terapéuticos son preparados de
caballos por inoculación de venenos completos. Sin embargo, los venenos de serpientes
contienen una amplia mezcla de componentes farmacológicamente activos. Aunque, no todos
los componentes son igualmente importantes para la producción de anticuerpos. Por lo tanto,
la identificación de las proteínas con capacidad inmunógenica, y además neutralizante,
pueden ayudar a definir los inmunógenos, para el desarrollo de antivenenos más eficientes.
Sin embargo, no siempre se cuenta con las cantidades deseadas de inmunógenos y es por esto
que el uso de proteínas recombinantes para ser utilizadas como inmunogénos se vuelve
importante.
Los objetivos principales en este trabajo fueron identificar y caracterizar
bioquímicamente al menos una proteasa y una fosfolipasa del veneno de Bothrops
ammodytoides, y expresarlas de forma heteróloga, para generar anticuerpos con capacidad
de reconocimiento hacia dichas proteínas nativas.
Durante el desarrollo del trabajo se logró la clonación y la expresión de tres enzimas:
PLA2, serinoproteasa, así como de una metaloproteasa utilizando el vector pQE30. La
actividad biológica de las enzimas recombinantes fue de baja a nula; esto debido, a que no
logró obtenerse un plegamiento satisfactorio de dichas enzimas. Esto es, la purificación de
cada una de las enzimas mostró varias fracciones con masas moleculares similares, indicando
indirectamente que cada fracción correspondía a un plegamiento diferente.
Independientemente de estos resultados, se inmunizaron animales con el fin de obtener
anticuerpos que reconocieran a las enzimas nativas.
Se obtuvieron IgG de caballos inmunizados con la PLA2 y serinoproteasa
recombinante, siendo la PLA2 recombinante la que mejores títulos generó. Los anticuerpos
anti-fosfolipasas recombinante redujeron la actividad de fosfolipasa y hemolítica en el
veneno de Bothrops ammodytoides.
Por otro lado, se generaron anticuerpos de conejo inmunizados contra las tres
proteínas recombinantes. Igualmente se encontró que la PLA2 y serinoproteasa
recombinantes fueron mejores inmunógenos para la producción de anticuerpos en conejos.
Como resultado general, se obtuvieron anticuerpos con poco reconocimiento a los venenos
completos del género Bothrops, pero se concluye que se pueden obtener anticuerpos de
proteínas recombinantes tanto en caballos como en conejos que reducen las actividades
enzimáticas de venenos de Bothrops. | es_MX |
dc.description | Abstract
In the American continent, the viperids ("vipers") are responsible for most of the
ophidic accidents, mainly by the Bothrops and Crotalus genera 1. Antibodies used for
antivenoms toward therapeutic purposes are built in horses by inoculation of complete
poisons. However, snake venoms contain a broad mixture of pharmacologically active
components; at the same time, not all the components are equally important for the
production of antibodies. Thus, the identification of proteins with immunogenic capacity,
and also neutralizing, can help define immunogens, for the development of more efficient
antivenoms. Nevertheless, the amounts of immunogens are not always available, and this is
the reason for using recombinant proteins as immunogens.
Our porpoises in this work were to identify and biochemically characterize, at least
one protease and one phospholipase from Bothrops ammodytoides snake venom.
Subsequently, obtain them in a recombinant way, in order to generate antibodies with
recognition capacity towards their native proteins.
During the development of the work, the cloning and expression of three enzymes
were achieved: PLA2, serine protease, as well as a metalloprotease using the pQE30 vector.
The Biological activity of the recombinant enzymes was low to none; mainly due to
inappropriate folding of those enzymes. After enzymes’ purification, we could find some
different fractions with similar molecular masses, indicating indirectly, that each fraction
corresponded to a singular folding. Regardless of these results, animals were immunized in
order to obtain antibodies that recognize native enzymes.
IgG from immunized horses with recombinant enzymes (PLA2 and serine protease)
were obtained, being the PLA2 the one that better titles generated. Anti-phospholipases
antibodies reduced hemolytic activity in Bothrops ammodytoides venom.
On the other hand, rabbit antibodies were obtained when the animal was immunized
with all three recombinant proteins. It was also found that recombinants PLA2 and serine
protease were better immunogens for the production of antibodies in rabbits. As a general
result, antibodies were obtained but with little recognition toward the complete poisons of
the genera Bothrops; however, we could obtain antibodies from recombinant proteins as well
in horses as in rabbits, and those antibodies are able to reduce the enzymatic activities of
