| dc.description | RESUMEN El objetivo del presente trabajo ha sido desarrollar y optimizar la formulación de películas mucoadhesivas con Metronidazol, combinando excipientes y propiedades tecnológicas de éstos, para obtener propiedades físicas de la formulación idóneas en base a las características de desempeño preestablecidas. Se realizó una búsqueda bibliográfica recabando información acerca de los mejores polímeros para cumplir con los objetivos establecidos, de igual manera se hicieron algunas pruebas con los polímeros para seleccionar los adecuados junto con los disolventes idóneos para mantener las propiedades requeridas para la formulación. Se realizó un diseño experimental con el programa estadístico Statgraphics, mediante el cual se fabricaron formulaciones en orden aleatorio y fueron evaluados en espesor, resistencia al plegado, porcentaje de hinchamiento, tiempo de desintegración de la película, tiempo de disolución de la película, fuerza de adhesión, para posteriormente realizar una optimización en base a las variables de respuesta estudiadas. La película mucoadhesiva optimizada fue evaluada para conocer el contenido de fármaco por cm2, la fuerza de mucoadhesión, de igual manera a estas películas se les evaluó la estabilidad durante un lapso de un mes a una temperatura de 40 ºC, obteniendo que éstas se mantenían estables sin crear ningún tipo de interacción fármaco-excipiente. Las películas obtenidas tuvieron una liberación inmediata, en una prueba de liberación in vitro, debido a que a los primeros 5 minutos liberaron el 100% del fármaco. También las películas fueron sometidas a pruebas microbiológicas para observar su efectividad de inhibición frente a cepas de Candidas, las cuales han sido reportadas en una alta prevalencia en infecciones vaginales obteniendo que las películas mucoadhesivas tienen una mayor eficacia que el fármaco en solución, su efectividad es comparable al Fluconazol, fármaco que es reportado como control por el CLSI (Clinical & Laboratory Standards Institute). | es_MX |
| dc.description | ABSTRACT The objective of this work has been developed and improved the formulation of mucoadhesive films with metronidazole, combining excipients and technological analysis properties, to obtain suitable physical properties of the formulation based on the pre-established performance characteristics. A bibliographic search was carried out, gathering information about the best protocols to meet the stated objectives. Similarly, some tests were carried out with the polymers to select the requirements together with the suitable solvents to maintain the properties required for the formulation. An experimental design was made with the statistical program, by which formulations were manufactured in random order and evaluated in thickness, resistance to folding, porcentage of swelling, time of disintegration of the film, concentration time of the film, strength of adhesion, to subsequently perform an optimization based on the response results studied. The optimized mucoadhesive film was controlled to know the content of the drug per cm2, the strength of mucoadhesion, in the same way, these films were evaluated for stability over a period of one month at a temperature of 40 ºC, obtaining that they remain stable without create any kind of excipient drug interaction. The films obtained had an immediate release, in an “in vitro” release test, because the first 5 minutes released 100% of the drug. The films also sometimes went to microbiological tests to observe their inhibition treatment against Candidas strains, which have been reported in a high prevalence in vaginal infections, obtaining the mucoadhesive films have a greater efficacy than the medicine in solution, their problem.
