dc.descriptionRESUMEN. En la producción de glicoproteínas como el interferón gamma (IFN-γ), la elección del sistema de expresión es crucial, porque influye en la calidad y cantidad de la proteína recombinante. Las células de mamíferos son los huéspedes preferidos para la producción de estas proteínas, ya que generan complejas modificaciones postraduccionales, pliegan correctamente la proteína y son capaces de segregar el producto en el medio de cultivo. El éxito generalizado de la línea de células CHO se debe a su incomparable adaptabilidad, generalmente utilizada en la expresión estable o constitutiva de las glicoproteínas, ya que estos sistemas permiten caracterizar la función y estructura de proteínas recombinantes complejas. Bajo este tenor dentro del grupo de trabajo se han generado sistemas transitorios utilizando células de mamífero como hospederos para producir la proteína interferón gamma dentro de los cuales con el sistema HEK293/AD5-IFN- se optimizó la expresión de la proteína obteniendo así 19.2 pg/mL, siendo esta la concentración más alta obtenida en un sistema transitorio. Por ello, en el presente trabajo se pretende generar una línea celular CHO-S que exprese de manera estable interferón gamma en altas concentraciones y en forma de homodímero glicosilado, así como realizar la caracterización tanto de la línea celular como de la proteína producida. Con el fin de lograr este objetivo se realizó primeramente la construcción del vector de expresión pcDNA3/IFN-γ, Durante el análisis de la secuenciación de la construcción del plásmido se corroboró que el transgén insertado corresponde con la proteína de interés (IFN-γ). Para la generación del sistema de expresión estable se realizó una selección policlonal obteniendo dos minipools con producciones bajas de interferón gamma. Seguido de la selección clonal para obtener dos clonas con producción alta de IFN-γ expresando más de 1000 pg/mL de manera constante el transgén de interés por más de 200 generaciones desde su transfección, siendo considerablemente mayores comparadas con el sistema el transitorio HEK293/AD5-IFN- el cual produce 19.22 pg/mL con lo cual se corrobora la hipótesis planteada en este proyecto.es_MX

During the production of glycoproteins such as gamma interferon (IFN-γ), the choice of the expression system is crucial, as it has a great influence on the quality and quantity of the recombinant protein. Mammalian cells are the preferred hosts to produce these proteins, as they generate complex post-translational modifications, correctly fold the protein and can secret the product into the culture medium. The widespread success of the CHO cell line is due to its incomparable adaptability, generally used in the stable or constitutive expression of glycoproteins, as these systems allow the characterization of the function and structure of complex recombinant proteins. In this sense, the working group has generated transitional systems using mammalian cells as hosts to produce the interferon-gamma protein. With the HEK293/AD5-IFN-γ system, the protein expression was optimized to obtain 19.2 pg/mL, which is the highest concentration obtained in a transitional system. Therefore, the present work aims to generate a CHO-S cell line that stably expresses gamma interferon in high concentrations and in the form of glycosylated homodimer, as well as to perform the characterization of both the cell line and the produced protein. In order to achieve this objective, the expression vector pcDNA3/IFN-γ was first constructed. During the analysis of the plasmid construction sequencing, it was corroborated that the inserted transgene corresponds to the protein of interest (IFN-γ). For the generation of the stable expression system a polyclonal selection was performed obtaining two minipools with low gamma interferon productions. Following the clonal selection to obtain two clones with high production of IFN-γ expressing more than 1000 pg/mL constantly the transgene of interest for more than 200 generations since its transfection, being considerably higher compared to the system the transitory HEK293/AD5-IFN-γ which produces 19.22 pg/mL which corroborates the hypothesis raised in this project.es_MX
